ELISA is a golden standard immunoassay technique used for determination of specific proteins, mostly antibodies. Using the simple principle of antibody-antigen bond, it gives us the possibility to measure the concentration of virtually any analyte.

Technology How it works?
Sandwich ELISA
Antigen – antibody (from sample) – antibody with HRP (conjugate) format.
Antigens are native, recombinants and or mix of the two. Specific antibodies from the sample bind with antigens during first incubation. All other non-specific antibodies are then washed away.
In the second step, secondary antibodies (which bind to human antibodies from the sample) are added. These antibodies are conjugated with horseradish peroxidase, an enzyme that converts substrate to color product. Reaction is stopped with acidic solution. The intensity of the color is proportional to the concentration of target antibodies from the sample. The color intensity is measured by a photometer at 450 nm.
This is the most used ELISA method across our portfolio.
Capture ELISA
Antibody (capture) – antibody (from sample) – antigen bound to monoclonal antibody conjugated with HRP (conjugate-tracer) format.
Microplate is coated with antibodies specific to human antibodies in the sample. Unbound antibodies are then washed away.
Afterwards, a tracer (specific antigen to targeted antibody bound to another antibody conjugated with HRP) is added. HRP then converts substrate to a color product.
This type of ELISA is used in EIA Toxoplasma kits in IgA, IgE and IgM antibody class to achieve higher sensitivity and specificity.
Antibody Avidity
Avidity is the strength of the non-covalent bond between antigen and antibody. It is typically used for IgG antibodies.
Low avidity antibodies are present during the acute phase of the first infection. They mature to high avidity antibodies over time. High avidity antibodies are typical for reinfections.
With the addition of urea solution, hydrogen bonds within antibody-antigen bonds are disrupted in the case of low avidity antibodies. Strong avidity antibody-antigen bonds persist even after the addition of urea.
Avidity is measured in CMV, EBV VCA, HSV, Measles, Rubella, TBEV, Toxocara and Toxoplasma kits. The avidity solution is always included in the kit and is not required to be bought separately.

Dynex Agility® system can process virtually any conventional, ready-to-use ELISA kit, that is meant to be processed on an open platform ELISA instrument. Kits are divided into different categories, depending on the compatibility:
- Gold kit – All TestLine EIA kits, ready-to-use SmartKit®.
- Silver kit* – Kits with reagent bottles that fit into a SmartKit®, but are not supplied as SmartKits®.
- Bronze kit* – Kits with reagents that need to be transferred to SmartKit® compatible bottles.
*For Silver and Bronze kit, SmartKit® inserts are not part of the kit and must be ordered separately.
